Target Information
LATS2 is a serine/threonine protein kinase belonging to the LATS tumor suppressor family. It interacts with a negative regulator of p53 and functions in a positive feedback loop with p53 that responds to cytoskeleton damage, provoking centrosome/mitotic apparatus dysfunction. LATS2 plays an essential role in maintaining mitotic fidelity and genomic integrity, and regulates organ size control and tumor suppression by restricting proliferation and promoting apoptosis.
The pathway involves a kinase cascade wherein STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 with MOB1. This in turn phosphorylates and inactivates YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 inhibits its nuclear translocation, regulating genes important for cell proliferation, death, and migration. It negatively regulates G1/S transition by down-regulating cyclin E/CDK2 kinase activity.
