The BMA12 monoclonal antibody reacts with mouse CD100, also known as Sema4D. CD100/Sema4D is a 150 kDa transmembrane protein constitutively expressed on T cells and involved in B cell activation through interaction with CD72. This interaction reduces the negative signaling mediated by CD72. CD100 is strongly expressed on resting T cells and weakly on B cells, with expression upregulated upon activation. Expression has been observed in spleen, thymus, and lymph nodes.

Target Information

Semaphorins are neuronal chemorepellents guiding neuron development. CD100 (Semaphorin 4D) is a 150 kDa homodimer that induces B cell aggregation and enhances viability in vitro. It modulates CD40-CD40L signaling by promoting B cell aggregation and survival, while downregulating CD23 expression. CD72 acts as a receptor for CD100, regulating B cell signaling. CD100 is expressed on resting and PHA-stimulated T cells, weakly on NK cells, EBV-transformed B cells, monocytes, and tumor/peripheral blood T cell lines, with higher expression on activated T and B cells.
