Target Information

Thioredoxins (Trx) are small, multifunctional proteins with oxidoreductase activity found in nearly all living cells. They contain a redox-active disulfide/dithiol group within the conserved Cys-Gly-Pro-Cys active site. The two cysteine residues can form intramolecular disulfide bonds when oxidized. Reduction of the active site disulfide in oxidized Trx is catalyzed by Trx reductase using NADPH as an electron donor. Reduced Trx acts as a hydrogen donor for ribonucleotide reductase, an essential enzyme for DNA synthesis, and functions as a general protein disulfide reductase involved in growth and redox regulation. Targets for Trx reduction include protein disulfide-isomerase (PDI) and transcription factors such as p53, NF-κB, and AP-1. Trx also contributes to H₂O₂ removal, especially when coupled with methionine sulfoxide reductase or peroxiredoxin isoforms.
