Product Specific Information
This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily. It contains an ITIM domain and is expressed on subsets of gamma-delta T cells, NK (CD56dim), CD8+, and CD4+ T cells.
KLRG1 is primarily expressed by effector/memory phenotype cells that are short-lived but capable of immediate effector function. Cadherin-E, -N, and -R are ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation.
The percentage of KLRG1 positive cells can vary depending on antigen experience. Clones 13F12F2 and 13A2 recognize a similar epitope based on cross-blocking studies.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (1.0 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is intended for use on cytometers equipped with a 355 nm ultraviolet laser.

Buffer Recommendation:

Target Information
SIGLEC5 is a putative adhesion molecule mediating sialic-acid dependent binding to cells. It binds both α2,3- and α2,6-linked sialic acids. The recognition site may be masked by cis interactions with sialic acids on the same cell surface.
Expressed by monocytic/myeloid lineage cells, SIGLEC5 is found at high levels in peripheral blood leukocytes, spleen, and bone marrow, and lower levels in lymph node, lung, appendix, placenta, pancreas, and thymus.
It is expressed by monocytes and neutrophils but absent from leukemic cell lines representing early myelomonocytic differentiation.
