Target Information
This gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which act as inhibitors of CDK4 kinase. Another transcript includes an alternate first exon located 20 Kb upstream, encoding the ARF protein, which stabilizes p53 by interacting with and sequestering MDM2, an E3 ubiquitin-protein ligase responsible for p53 degradation. Despite structural and functional differences, the CDK inhibitor isoforms and ARF protein share a role in cell cycle G1 control. This gene is often mutated or deleted in various tumors and is recognized as a key tumor suppressor gene.
