Product Specific Information

The SHLR17 monoclonal antibody reacts with human interleukin (IL)-17F, a homodimeric 34 kDa cytokine of the IL-17 family. IL-17F and IL-17A are preferentially produced by Th17 cells and play overlapping but distinct roles in inflammation. IL-17F binds to the IL-17RA/IL-17RC receptor complex, inducing secretion of pro-inflammatory cytokines and recruiting neutrophils.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume). Cell count per test may range from 10⁵ to 10⁸ cells.

Fluorochrome Information:
Brilliant Ultra Violet™ 563 (BUV563) emits at 564 nm and is suitable for UV (355 nm) laser-equipped cytometers. Protect from light to prevent photo-induced oxidation.

Buffer Recommendations:
When using multiple polymer dyes, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce non-specific interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).

Excitation / Emission / Laser:
350 nm / 564 nm / Ultraviolet Laser.

Target Information

IL-17F (Interleukin 17F, CTLA-8) is a cytokine produced by activated T cells, mast cells, and basophils. It plays a key role in allergic airway inflammation and induces cytokines, chemokines, and adhesion molecules in epithelial and endothelial cells. IL-17F forms homodimers or heterodimers with IL-17A and binds IL-17R to recruit monocytes and neutrophils. It stimulates TNFα, IL-1β, IL-6, and IL-8 expression and is implicated in chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis. IL-17F also inhibits angiogenesis and induces IL2, TGFB1, and MCP-1 production in endothelial cells.
