Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (low-affinity Fc gamma RIII). CD16 includes two isoforms, FcγRIIIA and FcγRIIIB.

• FcγRIIIA: expressed on monocytes, macrophages, and NK cells
• FcγRIIIB: expressed on neutrophils as GPI-anchored form

Binding of IgG activates signal transduction pathways leading to ADCC, phagocytosis, cytokine release, and antigen presentation.
3G8 recognizes the same epitope as CB16 but binds a distinct epitope from B73.1.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg)/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails prior to combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates use Phiton technology based on novel nucleic acid dye structures for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD16 encodes a receptor recognizing the Fc portion of immunoglobulin G, involved in immune complex clearance and cellular cytotoxicity.

• Gene: FCGR3A (chromosome 1)
• Expressed on NK cells (transmembrane anchored) and neutrophils (PI-anchored via FCGR3B).
• Mutations associated with Immunodeficiency 20, viral infection susceptibility, lupus, and neonatal neutropenia.
• Alternative splicing yields distinct isoforms.
