Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (FcγRIII).
CD16 exists as two isoforms: FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, NK cells
• FcγRIIIB: Expressed on neutrophils as a GPI-anchored form

Binding of IgG activates signal transduction, leading to ADCC, phagocytosis, cytokine release, and antigen presentation.
3G8 clone recognizes the same epitope as CB16 but distinct from B73.1.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (0.6 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

Important Usage Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
  Add 5 µL per sample for optimal staining with NovaFluor conjugates.
  For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and cell-mediated cytotoxicity.
The gene FCGR3A (chromosome 1) is expressed on NK cells via transmembrane peptide anchoring, while FCGR3B is expressed on neutrophils via phosphatidylinositol linkage.
Mutations may cause immunodeficiency 20 and susceptibility to viral infections or autoimmune diseases such as systemic lupus erythematosus.
