Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (Fc gamma RIII), which exists as two isoforms: FcγRIIIA and FcγRIIIB.

  • FcγRIIIA: Expressed on monocytes, macrophages, and NK cells.
  • FcγRIIIB: Found on neutrophils (GPI-anchored form).

Expression patterns differ among primates; for example, CD16 is not found on neutrophils in macaques and baboons but is detected in sooty mangabeys.
Binding of IgG activates ADCC, phagocytosis, cytokine release, and antigen presentation.

3G8 recognizes the same epitope as CB16 but differs from B73.1 clone.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells (5 µL (0.4 µg)/test).
Cell number: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
    • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and cell-mediated cytotoxicity.
The gene (FCGR3A) is similar to FCGR3B on chromosome 1.

  • FCGR3A: Expressed on NK cells (transmembrane anchored).
  • FCGR3B: Expressed on neutrophils (PI-anchored).

Mutations are linked to immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and neonatal neutropenia.
Alternative splicing produces multiple isoforms.
