Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

  • In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation and transcription of B cell-specific genes such as Prdm1 (Blimp1).
  • In T cells, IRF4 is implicated in Treg, Th2, Th9, and Th17 cell development and is upregulated upon activation.
  • IRF4 also plays roles in macrophage polarization and is upregulated in several hematologic malignancies.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested using human peripheral blood cells and the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).

Recommended use ≤ 1.0 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ per test. Antibody titration is advised for optimal performance.

Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm; use with a 405 nm violet laser. Check instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Not compatible with methanol-based fixation.

Target Information

The IRF (interferon regulatory factor) family of transcription factors features a unique tryptophan pentad repeat DNA-binding domain. IRFs regulate interferons in response to viral infection and control interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates TLR signaling, essential for innate and adaptive immune responses. Chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may cause multiple myeloma. Alternatively spliced transcript variants exist.
