Product Specific Information

Description:
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are identified ligands for PD-1. The J43 antibody inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo administration of J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes.

• Suitable for flow cytometric analysis.

Applications Tested:

• Tested on mouse splenocytes.
• Recommended ≤ 0.5 µg per test (final volume 100 µL).
• Cell number: 10⁵–10⁸ cells/test (empirically determined).
• Careful titration recommended for optimal performance.

• For use with cytometers equipped with a 405 nm (violet) laser.

Target Information

T-cell activation is regulated by inhibitory receptors such as PD-1 (PDCD-1). PD-1 binds ligands PDL-1 and PDL-2 on antigen-presenting cells to deliver inhibitory signals that limit immune activation. PD1-deficient mice show hyperactivated immune systems and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
