Product Specific Information

The monoclonal antibody 44.189 reacts with human CD336, also known as NKp44 or NCR2. CD336 is a 44-kDa transmembrane glycoprotein in the natural cytotoxicity receptor (NCR) family and contains an ITIM domain in its cytoplasmic tail. Expression and NK cell activation require association with DAP12, an ITAM-containing transmembrane accessory protein.

Originally thought to be expressed only on IL-2-activated NK cells and some γδ T lymphocytes, CD336 is also found on IL-3-stimulated plasmacytoid dendritic cells, αβ T cells in the small intestinal epithelium, and T cells in cord blood. CD336 participates in innate NK cell responses against viral infection and tumor proliferation.

The 44.189 antibody facilitates CD336 crosslinking and is validated for flow cytometric analysis of stimulated normal human peripheral blood cells. Recommended usage is 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell numbers may vary from 10⁵ to 10⁸ cells/test.

eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure your instrument supports violet laser detection.
