Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells (and sometimes B cells) and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
While CD28 provides costimulatory signals, CTLA-4 acts as an inhibitory receptor that suppresses IL-2 secretion and T cell proliferation. A large proportion of CTLA-4 is intracellular due to ER retention motifs, requiring intracellular staining for full detection.

The 14D3 antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Recommended use: 5 µL (0.03 µg) per test (100 µL final volume). Cell density: 10⁵–10⁸ cells/test.

For complete CTLA-4 detection, assess both surface and intracellular expression.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor primarily expressed on CD4⁺ T cells. It binds CD80 and CD86 with higher affinity than CD28, delivering inhibitory signals that downregulate T cell activation. Intracellular CTLA4 is also expressed in regulatory T cells, contributing to immune regulation.
