Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L (LFA-1 α chain). CD11a associates non-covalently with CD18 (integrin β2) to form LFA-1, an adhesion molecule involved in lymphocyte and granulocyte function. Its ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed by all leukocytes.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails or single-color controls.
• NovaFluor conjugates use Phiton™ technology for engineered fluorescent signatures with minimized spillover.
• Excitation: 552 nm Emission: 700 nm Laser: 561 nm (Yellow)
• Internal testing shows non-specific staining of B cells in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD11a (LFA-1 alpha, ITGAL) forms part of the LFA-1 (αLβ2 integrin) complex with CD18.
It mediates leukocyte adhesion via ICAM-1, -2, and -3, and participates in lymphocyte costimulatory signaling.
CD11a expression on leukocytes transitions from low- to high-affinity conformations upon cell stimulation, enabling tight ligand binding.
It plays roles in immune cell interaction, migration, differentiation, and T-cell effector function.
Blocking CD11a function is a therapeutic strategy for inflammatory diseases (e.g., Efalizumab, simvastatin-related modulation).
Two transcript variants encoding different isoforms have been identified.
Associated diseases include benign cephalic histiocytosis and leukocyte adhesion deficiency.
