Target Information
Glycogen synthase kinase-3 (GSK3)는 원래 glycogen synthase의 조절자로 알려진 protein kinase로, glycogen 대사에 중요한 역할을 합니다. 이후 다양한 세포 기능 — 단백질 합성, 세포 증식, 분화, 미세소관 조립 및 해체, 세포사멸 등 — 의 조절에도 관여하는 것으로 밝혀졌습니다.
GSK3는 독특한 기질 특이성을 가지며, GSK 인산화 부위의 C-말단에 phosphoserine 또는 phosphotyrosine이 존재할 때만 기질 인산화가 일어납니다. 두 가지 isoform(GSK3α, GSK3β)이 존재하며, 인산화에 의해 엄격하게 조절됩니다. GSK3β의 Ser9 (GSK3α의 Ser21)이 PKB에 의해 인산화되면 비활성화되며, 이는 성장 인자에 의한 kinase 억제의 주요 메커니즘입니다. 반면 Tyr216 (GSK3α의 Tyr279)의 인산화는 활성화를 유도합니다.
활성화된 GSK3β는 p53, c-Myc, c-Jun, HSF-1, cyclin D1 등 다양한 세포 단백질을 인산화할 수 있으며, Alzheimer’s disease 진행에 중요한 tau 단백질의 비정상적 인산화에도 관여합니다.
