• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: heat sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

Milk lipid globules from humans, cows, and rats contain a protein identified as adipocyte differentiation-related protein (ADFP). It is associated with the globule surface membrane material and was previously believed to be specific to adipocytes. ADFP (Adipophilin) is a major constituent of the lipid globule surface and is found in a detergent-insoluble complex containing butyrophilin and xanthine oxidase.
ADFP is expressed in a wide range of cultured cell lines, including fibroblasts, endothelial, and epithelial cells. In tissues, expression is restricted to specific cell types such as lactating mammary epithelial cells, adrenal cortex cells, Sertoli and Leydig cells, and hepatocytes showing steatosis in alcoholic liver cirrhosis.
ADFP may serve as a marker for identifying specialized differentiated cells containing lipid droplets and for diseases associated with fat-accumulating cells.
