Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II molecules and plays a key role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Tested: Flow cytometric analysis of mouse splenocytes (≤ 0.2 µg/test)

A test is defined as the amount (µg) of antibody that stains a cell sample in a final volume of 100 µL. Cell number may vary from 10^5 to 10^8 cells/test. Titrate for optimal performance.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD4 antigen is a co-receptor for MHC class II recognition and HIV binding. It is primarily expressed in T-helper lymphocytes, but also found in monocytes, macrophages, and dendritic cells. CD4 expression is seen in thymus, lymph nodes, tonsils, spleen, and other tissues. It functions in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may play a role in neuronal damage in immune-mediated CNS diseases.
