Product Specific Information
- For immunohistochemistry (PFA fixed), incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
This enzyme forms a dimer of two identical 70 kDa subunits attached to the plasma membrane via a glycosyl phosphatidylinositol linkage.
CD73 serves as a marker of lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
Other 5'-nucleotidase forms exist in the cytoplasm and lysosomes, differing from CD73 in substrate affinity, magnesium ion requirement, ATP activation, and phosphate inhibition.
The CD73 gene is located on chromosome 6q14–q21. Defects can cause joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding distinct isoforms have been identified.
