Target Information

The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator. The 20S core consists of four rings of 28 subunits (two alpha and two beta rings), while the 19S regulator includes a base with ATPase and non-ATPase subunits and a lid with up to ten non-ATPase subunits. Proteasomes are abundant in eukaryotic cells and degrade peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway.
The immunoproteasome variant processes class I MHC peptides. The PSMD2 gene encodes a non-ATPase subunit of the 19S regulator lid, which may also participate in the TNF signaling pathway through interaction with the TNF type 1 receptor. A pseudogene exists on chromosome 1.
