Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 is a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells, 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Use 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD4 antigen recognizes MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed in T helper cells, as well as monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 aids T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
