Product Specific Information
The L26 monoclonal antibody recognizes human CD20, a two-transmembrane spanning protein expressed on precursor and mature B lymphocytes. Expression is lost upon differentiation to plasma cells. CD20 levels are important in evaluating malignant B lymphocytes, including Non-Hodgkin’s lymphoma and various leukemias. CD20 plays a role in B cell differentiation and activation.
The L26 antibody binds to an epitope in the cytoplasmic domain, requiring tissue permeabilization for staining. Unlike antibodies recognizing the extracellular domain, L26 binding is not blocked by therapeutic anti-CD20 antibodies such as Rituximab.
Applications Reported: Immunohistochemical staining of FFPE tissue sections (IHC-P).
Applications Tested: IHC on FFPE human tonsil with low pH antigen retrieval. Titration recommended for optimal performance. Not validated for flow cytometry.
Filter Recommendation:
Use filters capturing 615 nm emission (e.g., Excitation 560/55, 585LP, Emission 645/75). Alexa Fluor™ 594 filter acceptable.

Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 associates with lipid rafts and interacts dynamically with the B-cell receptor (BCR) upon activation.
CD20 remains on the cell surface following antibody treatment, making it a useful target for therapeutic B-cell depletion. Dysfunctions are associated with Ms4a1-related common variable immune deficiency.
