Target Information
TREX2 is one of two major human 3′ to 5′ exonucleases required for checkpoint signaling after DNA damage. The related protein TREX1 is involved in ATR expression, and mutations in TREX1 are associated with Aicardi-Goutieres syndrome, chilblain lupus, and Cree encephalitis. TREX2 is ubiquitously expressed in all examined tissues.
X-ray crystallography studies of TREX2-single strand DNA complexes show that TREX2 binds DNA as a dimer, potentially displacing the second DNA strand and unzipping double-stranded DNA to feed the substrate into its active site. At least two isoforms of TREX2 are known.
