Background

The protein encoded by this gene belongs to the fibroblast growth factor receptor (FGFR) family, which shows high amino acid sequence conservation among members and across evolution. FGFR family members differ in ligand affinities and tissue distribution. A full-length FGFR-4 protein consists of:

  • Extracellular region with three immunoglobulin-like domains
  • Single hydrophobic membrane-spanning segment
  • Cytoplasmic tyrosine kinase domain

The extracellular domain interacts with fibroblast growth factors, triggering downstream signaling cascades that influence mitogenesis and differentiation. The gene structure contains 18 exons, unlike FGFR1–3 which have 19–20. Although alternative splicing occurs, there is no evidence that the C-terminal half of IgII is affected.
