Product Specific Information

For reconstitution, add 100 µL of distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (recommended: Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The transcription of many RNA polymerase II-dependent genes requires Spt3, a member of the S. cerevisiae SAGA complex.
Spt3 and Spt20 recruit TATA-box binding protein (TBP) to the core promoter, enabling TBP binding to SAGA-dependent promoters.
Null mutations in the Spt3 gene cause defects in sporulation, diploid filamentous growth, and haploid invasive growth, indicating its essential role in mating and developmental pathways.
At certain promoters (e.g., HO, HIS3, TRP3), Spt3 inhibits TBP binding, reducing transcription. This repression can be counteracted by GCN5, another SAGA complex member that promotes TBP/TFIIA complex formation via histone acetylation.
