This 2SX21R monoclonal antibody reacts with human CD360 (Interleukin-21 Receptor, IL-21R).
IL-21R forms a heterodimeric receptor complex with the common gamma chain to form a functional receptor for IL-21.
IL-21 signals through the IL-21R complex to activate the JAK/STAT pathway, promoting transcription of STAT1 and STAT3 responsive genes.

IL-21 is mainly produced by CD4+ T cells and mediates proliferation and differentiation of B cells, T cells, NK cells, dendritic cells, and other myeloid cells.
IL-21 and its receptor are implicated in chronic viral infections, autoimmunity, allergy, organ transplantation, and cancer.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).
• Suitable for cell counts ranging from 10⁵ to 10⁸ cells/test.

Fluorophore Information:

• PerCP-eFluor® 710 emits at 710 nm, excited at 488 nm (blue laser).
• Recommended filter: 710/50 bandpass (alternative: 695/40).
• Light-sensitive tandem dye — protect from light.

Fixation:

• Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).
• Clone-specific performance should be determined empirically.

Target Information

The IL-21 receptor is a type I cytokine receptor forming a heterodimer with the common gamma-chain, shared by IL-2 and IL-5 receptors.
It transduces IL-21 growth-promoting signals and regulates proliferation and differentiation of T cells, B cells, and NK cells.
Activation leads to signaling via JAK1, JAK3, STAT1, and STAT3.
Knockout studies suggest involvement in immunoglobulin regulation.
Three alternatively spliced transcript variants encoding the same protein have been described.
