Product Specific Information

The monoclonal antibody 44.189 reacts with human CD336 (NKp44/NCR2), a 44-kDa transmembrane glycoprotein of the natural cytotoxicity receptor (NCR) family containing an ITIM domain. CD336 expression and NK cell activation require association with DAP12, an ITAM-containing accessory protein.

Originally thought to be expressed only on IL-2-activated NK cells and some γδ T lymphocytes, CD336 has also been identified on IL-3-stimulated plasmacytoid dendritic cells, αβ T cells in the small intestinal epithelium, and T cells in cord blood. It plays a role in the innate immune response against viral infections and tumor proliferation.

The 44.189 antibody facilitates CD336 crosslinking and is validated for flow cytometric analysis. It is pre-titrated and tested using IL-2-stimulated normal human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:

• PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
• Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
• Ensure instrument compatibility before use.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD336 (NKp44/NCR2) is a natural cytotoxicity receptor involved in NK cell-mediated immune responses. It associates with DAP12 for activation signaling and is expressed on IL-2-activated NK cells, subsets of T lymphocytes, and dendritic cells. It contributes to antiviral defense and tumor suppression mechanisms.
