Product Specific Information
The HIP1 monoclonal antibody reacts with the human CD42b molecule, a 135 kDa transmembrane protein also known as platelet gpIb.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of human platelets.
Use at 5 µL (0.5 µg) per test (final volume 100 µL; cell number 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells.
- For single-color controls, use 5 µL of CellBlox per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information
CD42b is a platelet activation marker involved in coagulation and aggregation. It interacts with extracellular matrix and adhesion molecules and is regulated by VEGF and PDGF. CD42b forms part of the GPIb-IX-V receptor complex, essential for platelet-rich thrombus formation. It binds to von Willebrand factor, thrombin, coagulation factors XI/XII, high molecular weight kininogen, TSP-1, integrin Mac-1, and P-selectin. Expression occurs on platelets and megakaryocytes. Mutations in CD42b are associated with Bernard-Soulier syndrome and platelet-type von Willebrand disease.
