Product Specific Information

The HIP1 monoclonal antibody reacts with the human CD42b molecule, a 135 kDa transmembrane protein also known as platelet gpIb.
Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of human platelets.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes.

This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation over previous CellBlox.
CellBlox Plus Blocking Buffer is required for optimal staining with NovaFluor conjugates.

Usage recommendation:

• Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells.
• For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD42b is a platelet activation marker involved in coagulation and platelet aggregation.
It interacts with extracellular matrix and adhesion molecules and is regulated by VEGF and PDGF.
CD42b forms part of the GPIb-IX-V receptor complex critical for platelet-rich thrombus formation by tethering platelets to thrombogenic surfaces.
It binds to von Willebrand factor (VWF), thrombin, coagulation factors XI and XII, high molecular weight kininogen, TSP-1, integrin Mac-1, and P-selectin.
CD42b is expressed on platelets and megakaryocytes in bone marrow.
Mutations in CD42b are associated with Bernard-Soulier syndrome and platelet-type von Willebrand disease.
