Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the αLβ2 integrin.
CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAM-1, ICAM-2, and ICAM-3.
It also functions in lymphocyte costimulatory signaling and is expressed on leukocyte plasma membranes in a low-affinity conformation. Upon stimulation by chemokines or other signals, CD11a adopts a high-affinity conformation that enables tight binding to its ligands.
CD11a is involved in immune cell interactions, tissue-specific localization, and regulation of cell differentiation, proliferation, and T-cell effector functions.
Blocking CD11a function with specific antibodies or small molecules is an important therapeutic approach for inflammatory diseases. For example, the humanized anti-CD11a antibody Efalizumab (Raptiva) interferes with T cell migration, while simvastatin binding to CD11a’s allosteric site leads to immunomodulation.
Two transcript variants encoding different isoforms of CD11a have been identified. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
