Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, serving as key regulators of cell death and stress response.
TRAF proteins share a conserved region enabling receptor and TRAF-TRAF interactions, activating distinct signaling cascades.
TRAF2 and TRAF3 bind to CD40, a receptor involved in B cell immunity, and are essential for NF-κB activation in mouse B lymphocytes.
TRAF2 and TRAF6 are required for CD40 signaling in non-hematopoietic cells.
TRAF2 also interacts with the lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
