Target Information

TRAIL (TNF-related apoptosis-inducing ligand) is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells while sparing most normal cells. TRAIL binds to several TNF receptor superfamily members, including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly TNFRSF11B/OPG.
The activity of TRAIL may be modulated by decoy receptors (TRAILR3, TRAILR4, OPG) that cannot induce apoptosis. Binding of TRAIL to its receptors triggers activation of MAPK8/JNK, caspase 8, and caspase 3. TRAIL is a type II membrane protein expressed in many human tissues and induces apoptosis and NF-κB activation similar to TNF and Fas ligand.
