Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, constituting about 10% of surface proteins in lymphocytes. It is absent in non-hematopoietic cells and tissues.
CD45 is crucial in lymphocyte development and antigen signaling, regulating Src-family kinases. Multiple isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45RA, one isoform, shows restricted expression among lymphoid cell subtypes. CD45 isoforms vary in membrane domain localization and expression depending on cell type and physiological state.
CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases, and modulates cytokine receptor pathways. It also influences cell survival, integrin-mediated signaling, DNA fragmentation during apoptosis, and various immunological functions.
