Product Specific Information
Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of surface proteins in lymphocytes.
CD45 is absent in non-hematopoietic cells and tissues. It plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45RA is an isoform with restricted expression among lymphoid cell subtypes. Isoforms vary in membrane localization and expression depending on cell type and physiological state.
CD45 regulates T- and B-cell receptor signaling, modulates JAK kinases in cytokine signaling, and influences cell survival, apoptosis, and immune functions.
