Store vial at −20°C prior to opening. Aliquot contents and freeze at −20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. Stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

This antibody is specific for human Pdcd4 protein phosphorylated at Ser457. BLAST analysis suggests cross-reactivity with Pdcd4 from human, mouse, rat, and Xenopus based on 100% homology with the immunizing sequence. Cross-reactivity with Pdcd4 from other sources has not been determined.

Target Information

Apoptosis, or programmed cell death, is essential for development, tissue turnover, and tumor formation. PDCD4 (Programmed Cell Death Protein 4) expression increases during apoptosis and encodes a tumor suppressor protein lost in several carcinomas (breast, colon, lung, prostate). PDCD4 inhibits the helicase activity of eIF4A and suppresses AP-1-mediated transcriptional activation. Akt kinase phosphorylates PDCD4, reducing its ability to interfere with AP-1 transactivation. Two isoforms of PDCD4 are known.
