Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule (32–34 kDa), expressed as CD8αβ heterodimers or CD8αα homodimers.
- Major expression: thymocytes, mature αβ TCR T cells (CD8αβ), γδ TCR T cells, intestinal IELs, dendritic cells (CD8αα)
- CD8 binds MHC class I and associates with p56lck for T cell development and activation.
Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
Recommended Reagent:
Each NovaFluor conjugate ships with CellBlox Blocking Buffer.
- Add 5 µL per sample for staining or single-color controls.
- Add to antibody cocktails before combining with cells.

Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein acting as a co-receptor for MHC class I molecules.
- Predominantly expressed on cytotoxic T cells (CD8αβ heterodimers).
- Also found on NK cells, monocytes, and dendritic cells (CD8αα homodimers).
- CD8-MHC-I interaction recruits Lck kinase, inducing lymphokine production and CTL activation.
- CTL differentiation enhanced by IL-2, IL-12, and TGF-β1.
- Plays a key role in immune response and pathogen/tumor clearance.
