Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 exhibits tyrosine phosphatase activity and is essential for lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated by flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (0.8 µg) per test (in 100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal performance.
  • Add 5 µL per sample to antibody cocktails prior to cell staining.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures with minimized spillover and spread effects.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins. It is absent in non-hematopoietic cells and tissues. CD45 plays a critical role in lymphocyte development, antigen receptor signaling, and regulation of Src-family kinases. Isoforms (such as CD45RA) arise from alternative splicing and differ in extracellular domains, influencing their localization and function. CD45 also modulates integrin-mediated signaling, apoptosis, and cytokine receptor pathways.
