Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except erythrocytes and platelets. The cytoplasmic portion exhibits tyrosine phosphatase activity important for lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells; use 5 µL (0.8 µg) per test (100 µL total volume). Cell number range: 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of surface proteins in lymphocytes). Absent in non-hematopoietic tissues, CD45 regulates Src-family kinases, T/B-cell receptor signaling, and cytokine receptor pathways. Multiple isoforms arise via alternative splicing, differing in extracellular domains. CD45 also modulates integrin-mediated signaling, apoptosis, and immune functions.
