Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. It plays a critical role in lymphocyte activation through its tyrosine phosphatase activity.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: For use in flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test (100 µL final volume).

Cell number range: 10⁵–10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved performance over the original CellBlox.
Add 5 µL per sample before combining with cells for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures and reduced spillover/spread.

Target Information

CD45 (Leukocyte Common Antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent on non-hematopoietic cells and tissues.

CD45 is essential for:

• Lymphocyte development and antigen signaling
• Regulation of Src-family kinases
• Modulation of cytokine receptor signaling via JAK kinases
• Cell survival through integrin-mediated pathways
• Apoptosis and immunological function regulation

Isoforms (e.g., CD45RA) differ by alternative splicing and cell-type-specific expression.
