Product Specific Information

This antibody recognizes a conformationally-dependent extracellular epitope of CD8, a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system.
CD8 is a disulfide-linked dimer and exists as a CD8 alpha/alpha homodimer or CD8 alpha/beta heterodimer (each monomer approx. 32–34 kDa).
The antibody does not react with formaldehyde-fixed cells and is negative in Western blotting applications.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays an integral role in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
In contrast, CD4 serves as a co-receptor for MHC class II molecules.
CD8 is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Upon MHC-I/peptide complex binding, CD8 recruits the Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
