SIRT3는 효모 Sir2 단백질과 상동성을 가진 sirtuin 단백질 패밀리의 구성원입니다. 이 단백질들은 sirtuin core domain을 가지며 네 가지 클래스로 분류됩니다. 인간 sirtuin의 기능은 아직 완전히 규명되지 않았으나, 효모 sirtuin은 유전자 발현의 후성유전적 조절과 rDNA 재조합 억제에 관여하는 것으로 알려져 있습니다. 연구에 따르면, 인간 sirtuin은 세포 내 조절 단백질로서 mono-ADP-ribosyltransferase 활성을 가질 수 있습니다.
