Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
• Recommended use: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Conjugates:
Based on Phiton technology using novel nucleic acid dye structures, engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD4 Antigen:

• Involved in recognition of MHC class II molecules and co-receptor function for HIV.
• Expressed primarily in T helper lymphocytes, and also in monocytes, macrophages, and dendritic cells.
• Detected in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut.
• Plays a role in T-cell activation via association with T-cell receptor complex and kinase Lck.
• May mediate neuronal damage in infectious and immune-mediated CNS diseases.
• Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
