Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed on most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. The SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells.
Use 5 µL (0.1 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample or per antibody cocktail before mixing with cells.

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV receptor. It is primarily expressed on T helper lymphocytes and at lower levels on monocytes, macrophages, and dendritic cells. Expression is found in thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut. CD4 participates in T-cell activation via association with the T-cell receptor complex and Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known.
