Intercellular Adhesion Molecule-1 (ICAM-1) is a member of the immunoglobulin supergene family and functions as a ligand for the Lymphocyte Function-Associated Antigen-1 (LFA-1), an αβ complex belonging to the leukocyte integrin family of cell-cell and cell-matrix receptors.

ICAM-1 is a single-chain glycoprotein (~55 kDa core) expressed on various non-hematopoietic cells (e.g., vascular endothelial, thymic epithelial, fibroblasts) and hematopoietic cells (e.g., macrophages, activated T-lymphoblasts, germinal center B cells, dendritic cells).

Expression of ICAM-1 is inducible by inflammatory mediators such as IL-1, TNF, and IFN-γ, correlating with lymphocyte infiltration into inflammatory lesions. It is an early and strong marker of inflammation, often preceding HLA-DR expression.

Elevated soluble ICAM-1 (sICAM-1) levels are associated with acute renal graft rejection, insulin-dependent diabetes mellitus, anterior uveitis, and allergic inflammation.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein with five immunoglobulin-like domains, a transmembrane region, and a cytoplasmic tail. It has seven potential N-linked glycosylation sites and shares sequence homology with ICAM-2 (CD102) and ICAM-3 (CD50).

ICAM-1 binds to integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1) and serves as a receptor for Rhinovirus. It is expressed by activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils. Expression increases during inflammation and is also found in some carcinoma and melanoma cells. Soluble ICAM-1 is detectable in plasma and elevated in various inflammatory conditions.
