Description:
The MBSA43 mouse monoclonal recombinant antibody reacts with human TIGIT (T cell immunoreceptor with Ig and ITIM domains), also known as VSTM3 or WUCAM. TIGIT is a 26 kDa protein in the poliovirus receptor (PVR) family. Expression occurs on NK cells, regulatory T cells, follicular helper T cells, memory CD4+ and CD8+ T cells, but not on B cells or naive CD4+ T cells. TIGIT expression may increase upon activation of naive CD4+ T cells.

TIGIT interacts with PVR and PVR-like family members such as CD155, mediating NK and T cell interactions with antigen-presenting cells, fibroblasts, and endothelial cells expressing these proteins.

MBSA43 is reported to cross-react with Rhesus macaque.

Applications Tested:
Validated for flow cytometric analysis of human PBMCs. Recommended usage ≤ 0.25 µg per test (100 µL final volume). Cell number per test should be optimized (10⁵–10⁸ cells/test). Antibody titration is advised for best performance.

Target Information

TIGIT binds with high affinity to PVR, leading to increased IL10 and decreased IL12B secretion, and suppresses T cell activation by promoting mature immunoregulatory dendritic cell generation.
