Target Information

The 26S proteasome is a multicatalytic proteinase complex composed of two major structures: a 20S core and a 19S regulator.

  • The 20S core contains four rings of 28 non-identical subunits: two rings of seven alpha subunits and two rings of seven beta subunits.
  • The 19S regulator includes a base (six ATPase and two non-ATPase subunits) and a lid (up to ten non-ATPase subunits).
    Proteasomes are widely distributed in eukaryotic cells and function in ATP/ubiquitin-dependent peptide cleavage within a non-lysosomal pathway.
    The immunoproteasome variant is essential for processing class I MHC peptides.
    The PSMC4 gene encodes one of the ATPase subunits, a member of the triple-A family with chaperone-like activity. This subunit interacts with nuclear hormone receptor family members and gankyrin, a liver oncoprotein. Two transcript variants encoding different isoforms have been identified.
