Description:
The 1D11 monoclonal antibody reacts with human NKG2D, a 42 kDa lectin-like molecule expressed by NK cells, γδ T cells, CD8+ T cells, and some CD4+ T cells. NKG2D forms complexes with DAP10 and can costimulate multiple NK activation receptors. Its counter-receptors MICA/MICB are expressed on various epithelial tumors (lung, breast, kidney, ovary, prostate, colon carcinoma). 1D11 blocks soluble MICA binding to γδ TCR T cell clones and inhibits lysis by these cells, inducing redirected lysis of Fc receptor-bearing P815 cells.

Applications Reported:
Reported for use in flow cytometry, immunoprecipitation, and immunohistochemical staining of frozen tissue sections, as well as inhibition assays. Functional Grade Purified 1D11 (Product #16-5878) is recommended for functional assays. Fluorochrome-conjugated 1D11 is recommended for flow cytometry.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.125 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration is advised for optimal performance.

Target Information

Natural killer (NK) cells are lymphocytes that mediate lysis of tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express several calcium-dependent (C-type) lectins involved in NK cell regulation. The NKG2 gene encodes a type II transmembrane protein with a C-type lectin domain, located within the NK gene complex containing multiple NK-specific lectin genes.
