Product Specific Information

For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL (Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress.
It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation.
p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest.
Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation.
p53 plays a vital role in preventing proliferation of damaged cells and acts as an anticancer agent.
Mutations in TP53 are common in human cancers and lead to loss of tumor suppressor function.
It is also implicated in aging, oxidative stress, and cognitive function (LTP).
Alterations of TP53 occur as somatic or germline mutations, including in Li-Fraumeni syndrome.
