Target Information

Immunophilins are soluble cytosolic receptors that bind immunosuppressant agents such as cyclosporin A (CsA) or FK506.

• Proteins binding FK506 are termed FK506 Binding Proteins (FKBPs).
• Proteins binding cyclosporin A are called cyclophilins (CyP).

CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium/calmodulin-dependent protein phosphatase involved in T-cell activation, explaining the immunosuppressive mechanism of CsA and FK506.
Immunophilins act as peptidyl prolyl cis-trans-isomerases (PPIase), accelerating protein folding by catalyzing slow steps in proline-containing protein rearrangement.
Within the cyclophilin family, CyPA (also known as CyP-18) is the most abundant and ubiquitous form found in all vertebrate tissues, including T-cells.
