Target Information
Diacylglycerol (DAG) functions as an intracellular activator of protein kinase C (PKC) and guanine nucleotide exchange factors, influencing various cell signaling pathways. To maintain cellular homeostasis, DAG levels are regulated by diacylglycerol kinases (DGKs), which phosphorylate DAG to phosphatidic acid.
- Type I DGKs (DGK-α, -β, -γ): Contain calcium-binding EF-hand motifs.
- Type II DGKs (DGK-δ, DGK-ι): Contain N-terminal pleckstrin homology (PH) domains.
- Type III DGK-ε: Contains no identifiable regulatory domains (64 kDa).
- Type IV DGKs (DGK-ζ, DGK-ι): Contain C-terminal ankyrin repeats.
- Type V DGK-θ: Contains three cysteine-rich domains and a PH domain.
