This 23dcdp monoclonal antibody reacts with the p19 subunit of human IL-23, a heterodimeric cytokine composed of the p40 and p19 subunits. IL-23 is closely related to IL-12, sharing the p40 subunit. The IL-23 receptor is heterodimeric, consisting of IL-12Rβ1 and IL-23R. IL-23R signaling activates the Jak/STAT pathway and induces RORγt expression, promoting Th17 cell maintenance and proliferation.

Dendritic cells and macrophages produce IL-23 in response to TLR2, TLR4, and TLR8 ligands, as well as Dectin-1 agonists. Expression levels vary depending on donor, kinetics, and stimulant type. Some studies suggest that the p19 subunit may exist independently of p40 association.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining by flow cytometry. Use 5 µL (0.06 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Target Information

IL-23 is a heterodimeric cytokine consisting of the IL-12 p40 subunit linked to p19. The p19 subunit alone is inactive. IL-23 interacts with IL-12Rβ1 and IL-23R, activating STAT4 signaling. It is secreted by activated dendritic cells and exhibits distinct biological activities from IL-12.
In mice, IL-23 induces proliferation of memory T cells and IL-17 production, while IL-12 does not. In humans, IL-23 promotes memory T cell proliferation and moderate IFN-γ production compared to IL-12.
